If you haven't had a raise since you've been there, I don't feel that a 10 or 15 percent raise would be out of the question. But then again, I don't know the financial situation of your company or their compensation policies. Make sure that your compensation is brought up during the conversation. If the boss doesn't bring it up, you may have to.

They both sound like rather different fields, seeing as your initial degree is in Biology, Enterprise in Biotechnology sounds the better bet. Also IT Application development is reasonably well paid but the IT industry as a whole is very crowded with lots of competition, even myself I am currently studying for a Degree in Computer Science and I know I will struggle to find a decent job in the field (in this country). Personally I'm looking at opportunities abroad.

I often ask how the position has become vacant? It's interesting if the previous employee just upped and left, or was fired etc etc. I'm stumped after that! Another one I find difficult is 'what are your biggest downfalls?' Hey, I don't have any ;-)
